ATRX is a member of the Snf2 family of helicase/ATPases, which contribute to the remodeling of the nucelosome structure in an ATP-dependent manner, and facilitate the initiation of transcription and replication. Structurally, ATRX contains a PHD zinc finger motif. ATRX is regulated throughout the cell cycle where it is differentially distributed within the nucleus. During interphase, ATRX predominately associates with the nuclear matrix, while during mitosis, ATRX localizes with condensed chromatin. At the onset of M phase, phosphorylation rapidly induces this redistribution of ATRX to the short arms of human acrocentric chromosomes, where it then specifically complexes with heterochromatin protein 1 thalassemia or ATRX syndrome. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. The LMO2 protein has a central and crucial role in hematopoietic development and is highly conserved. It has a particular function in normal and lymphatic endothelial cells involving the regulation of angiogenesis and lymph-angiogenesis. Immunohistochemical studies have also demonstrated expression of LMO2 in both normal germinal center B-cells and germinal center-derived B-cell lymphomas, including follicular lymphoma and diffuse large B-cell lymphoma. The use of anti-LMO2 is valuable as a tool in the identification of lymphomas of B-cell origin. This MAb recognizes a protein of ~30 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol. This antibody recognizes a protein of 30-33 kDa, which is identified as CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas.
